Interaction of polyomaviruses and host cells; mechanism involved in reactivation of JC virus by the immune system.
Research Summary
The focus of our research lies in the area of mechanisms of regulation of JC virus (JCV) replication and transcription during the course of infection of glial cells. In particular, alterations in cytokine profiles that are associated with immunosuppressive states may play a role in the reactivation of JCV from latency in progressive multifocal leukoencephalopathy (PML). Attention is being focused on the effects of factors produced by activated T-cells that are able to suppress JCV replication when supplied to glial cell cultures. These effects are mediated by the upregulation of expression of a protein designated GBP-i that binds to a GGA/C-rich region (GRS) encompassing the JCV origin of DNA replication (5112 to +4). We are currently engaged in purifying this protein for identification by mass spectrometry. Other research includes investigation of dysregulation of cell-cycle checkpoints and cellular DNA repair processes leading to the accumulation of DNA damage during JCV infection. Evidence points to the involvement of the JCV proteins T-antigen and agnoprotein and their interactions with cellular proteins such as p53, IRS-1, Rad51 and Ku70. Cellular DNA damage accrued by oligodendrocytes and astrocytes likely contributes to cytotoxicity in PML and viral destruction of areas of white matter within the brain that is characteristic of PML pathogenesis. The ability of JCV T-antigen and agnoprotein to dysregulate cell growth and cause DNA damage may also contribute to malignant progression in tumors that express these proteins.
