1 Intranuclear Immunization. Role: P.I. In collaboration with E.E. Henderson (HIV) and K.J. Soprano (RARα)
The purpose of this project is to develop a mechanism for the regulated expression of single chain antibodies (linked heavy and light chain variable fragments) in cells and the transport of such antibodies to the nucleus of the cell. A system of vectors has been constructed and a monoclonal antibody to HIV integrase has been successfully introduced and expressed blocking the replication of HIV. A library of monoclonal antibodies has been generated to retinoic acid receptor RARα. One of these has been cloned and will be introduced into the vector system to determine its effect on the cellular response to retinoids. Additional antibodies will also be cloned and tested in hopes of mapping retinoid response elements.
2. Angiogenesis. Role: Co-investigator. In collaboration with Dr. R.W. Colman, and Dr. I.M. Sainz
A monoclonal antibody directed against D5 of high molecular weight kininogen has been shown to inhibit neovascularization of the chicken chorioallantoic membrane. When injected into mice the hybridoma producing this antibody gives rise to tumors which are significantly smaller than those produced by control hybridomas. Vascular density in these tumors is also significantly less than controls and the tumors regress by 22 days post injection, as opposed to continued and rapid growth of control hybridomas. The kinetics and mechanisms of these findings are being investigated to determine whether there is potential for clinical application of this antibody.
3. Gene Therapy. Role: Co-investigator. In collaboration with Dr. R. Rolandelli, and Dr. J. Migaly
A role for TGF-β gene therapy in intestinal surgery is being investigated. Maintaining tissue viability following colon resectioning is a common problem. There is evidence that the cytokine TGF-β can improve tissue viability and healing following surgery. Currently the TGF-β gene is expressed in an adenovirus vector. Dr. Long has expertise in virology and molecular biology. He is involved in propagation of the vector, subcloning the TGF-β gene into a more suitable adeno-associated virus vector, and determining levels of TGF-β expression.
4. Generation of Monoclonal Antibodies to Opioid Receptors. Role: Co-investigator. In collaboration with Dr. T. Rogers
Monoclonal antibodies to opioid receptors will be generated. Attempts to generate monoclonal antibodies to opioid receptors by a number of investigators have been unsuccessful because the receptor proteins are recognized as self. To avoid this problem, knockout mice which do not express receptors will be immunized and used as a source of cells in generating hybridomas producing the appropriate antibodies.
