Research Interests:

Dr. Knight’s work focuses on using in vivo imaging to follow the behavior of molecules and cells in vivo, and to use functional imaging to learn more about physiology.

Major areas of work include:    Development of radiopharmaceuticals which target vascular receptors. One project is to design and evaluate a new diagnostic imaging test for imaging thrombi and emboli. The approach currently under evaluation is based on a radiolabeled peptide which recognizes a receptor on activated platelets. Another project is to evaluate radiolabeled peptides which bind to receptors in the blood vessels of tumors. The goals of this project are to locate a wide variety of solid tumors by imaging, monitor the effects of anti-angiogenic therapy, and eventually treat the tumors with targeted radiotherapy using a therapeutic radionuclide bound to the same peptide.

Assessment of gastrointestinal motility. Physiologic meals can be prepared containing a radiotracer, so that their transit through the GI tract can be detected and quantified by noninvasive imaging. The effects of drugs and other interventions on the motility of segments of the GI tract can be assessed quantitatively in vivo.

Radiolabeling of drugs to determine their behavior in vivo.   Drugs to be administered to the GI tract have been radiolabeled to assess their duration and extent of coverage of GI mucosa in various diseases such as peptic ulcer and colitis.
