Structure-function studies of prostaglandin E receptors. Prostaglandins are lipid mediators released from most cells. They are involved in numerous physiological and pathological functions including inflammation, fever, blood pressure regulation and kidney function. Prostaglandin E2 acts through binding to four subtypes of receptor designated EP1 through EP4. The laboratory studies the structure-function relationships of the receptors by site-directed mutagenesis and heterologous expression. Current work is focused on the EP3 subtype, which occurs as several isoforms. In efforts to understand the significance of the isoforms we have used the yeast-hybrid system to identify proteins that bind to specific isoforms. Studies are underway to determine the physiological significance of these interactions.
